Age-related differences in autism: The case of white matter microstructure

Abstract

Autism spectrum disorder (ASD) is typified as a brain connectivity disorder in which white matter abnormalities are already present early on in life. However, it is unknown if and to which extent these abnormalities are hard-wired in (older) adults with ASD and how this interacts with age-related white matter changes as observed in typical aging. The aim of this first cross-sectional study in mid- and late-aged adults with ASD was to characterize white matter microstructure and its relationship with age. We utilized diffusion tensor imaging with head motion control in 48 adults with ASD and 48 age-matched controls (30-74 years), who also completed a Flanker task. Intra-individual variability of reaction times (IIVRT) measures based on performance on the Flanker interference task were used to assess IIVRT-white matter microstructure associations. We observed primarily higher mean and radial diffusivity in white matter microstructure in ASD, particularly in long-range fibers, which persisted after taking head motion into account. Importantly, group-by-age interactions revealed higher age-related mean and radial diffusivity in ASD, in projection and association fiber tracts. Subtle dissociations were observed in IIVRT-white matter microstructure relations between groups, with the IIVRT-white matter association pattern in ASD resembling observations in cognitive aging. The observed white matter microstructure differences are lending support to the structural underconnectivity hypothesis in ASD. These reductions seem to have behavioral percussions given the atypical relationship with IIVRT. Taken together, the current results may indicate different age-related patterns of white matter microstructure in adults with ASD
Le trouble du spectre de l'autisme (TSA) se caractérise comme un trouble de la connectivité du cerveau dans lequel les anomalies de la substance blanche sont déjà présentes au début de la vie. Cependant, on ne sait pas si et dans quelle mesure ces anomalies sont câblés chez (âgés) les adultes avec TSA et comment cela interagit avec les changements de la substance blanche liés à l'âge comme observés dans le vieillissement typique. Le but de cette première étude transversale chez l'adulte d'âge moyen et tardif avec TSA était de caractériser la microstructure de la matière blanche et sa relation avec l'âge. Nous avons utilisé l'imagerie à tenseur de diffusion avec le contrôle du mouvement de la tête chez 48 adultes avec TSA et 48 adultes contrôles appariés selon l'âge (30-74 ans), qui ont terminé aussi une tâche Flanker. Les mesures de la variabilité intra-individuelle des temps de réaction (IIVRT) basées sur la performance de la tâche d'interférence Flanker ont été utilisées pour évaluer les associations de microstructure de la matière blanche IIVRT . Nous avons observé principalement l'augmentation moyenne et radiale de la diffusivité dans la microstructure de la matière blanche dans le TSA, en particulier dans les fibres à longue portée, qui ont persisté après la prise en compte du mouvement de la tête . Fait important, les interactions de groupe par âge a révélé une moyenne plus élevée liée à l'âge et de la diffusivité radiale dans le ASD, dans les fibres des voies de projections et d'associations. Des dissociations subtiles ont été observées dans les relations microstructure de la matière blanche IIVRT entre les groupes, avec le motif d'association de matière blanche IIVRT dans dans le TSA ressemblant à des observations dans le vieillissement cognitif. Les différences de microstructure de la substance blanche observées sont prêts à appuyer l'hypothèse de la sous-connectivité structurelle dans le TSA. Ces réductions semblent avoir des percussions comportementales étant donné la relation atypique avec IIVRT. Pris ensemble, les résultats actuels peuvent indiquer différents schémas liés à l'âge dans la microstructure de la matière blanche chez les adultes avec TSA.
